I just installed Theme Builder, but in Choose Layout under Configure Theme, there only have Default theme listed. Does the Theme Builder come with more than Default theme?

There are some inbuilt layouts within builder however you must manually import them through the 'import default layouts' button on the backup tab of builder administration (link on the left hand side of site admin)
